Appearance of Solution | What is happening to the cells/DNA? | |
After lysis buffer | Started off hydrophobic. It clumped together and sank to the bottom of the liquid, after shaking it a few times, it was more cloudy | The individual cells are being split apart by the detergent and salt to isolate the DNA. |
After alcohol added | The alcohol and the solution did not mix creating a small layer. | The solubility of the DNA is being affected by the alcohol. That’s why it stayed at the bottom. |
After alcohol mixed | After mixing, the tube was clear, at the bottom were little floaty string things. | The remaining cells dissolved and all that was left were strands of DNA that you could see with the naked eye. |
