Objective:
- to acquaint us of microscope operations by observing various types of bacteria
- observe and gain better knowledge of the many bacterial shapes, sizes, and structures in cultural yogurt and prepared slides
Materials:
- 1 microscope
- 1 oil immersion
- 1 plain active cultural yogurt
- 1 collection container’
- 1 toothpick’
- 1 slide of Bacteria Coccus Form
- 1 slide of Bacteria Spirillum
- 1 slide of Anabeana, Whole Mount
- 1 slide of Pencicillium with Conidia
- 1 slide of Yoghurt Bacteria
- 1 slide of Bacteria Bacillus Form
- 1 pair of Disposable Gloves
- 1 sheet of lens paper
- 1 blank slide
- 1 cover slip
Procedure for Exercise #1: Prepared Slides:
1. Prepare the microscope by cleaning the oculars and objectives
2. Put disposable gloves on to ensure that the slides do not get finger prints on them’
3. Retrieve desired slide
4. Place the slide on the stage under the clips to ensure that movement of the slide is disabled
5. Use the X and Y travel knobs in order to move the slide to find the best view
6. Rotate the nosepiece to the 40x objective, focus the specimen and be sure to record your observations by taking a photo
7. Put oil immersion on to the slides as the objectives are out of the way
8. Rotate the nosepiece to the 100x objective, focus the specimen (as well as you can) and be sure to record your observations by taking a photo
9. Repeat these 8 steps for the first 6 prepared slides
Procedure for Exercise #2: Bacteria Cultures in Yogurt
1. Open the container of plain active cultural yogurt, with a spoon, take out a tablespoon of yogurt and place it in a container
2. Let the yogurt set in a warm, dry place for 24 hours
3. Once the 24 hours have passed, use a toothpick and take a sample of the yogurt
